place the graticule in the microscope, then see the number of divisions which are coming between the tissue or cell part being observed (according to what asked), then remove the slide and place stage micrometer. see how many divisions of eye piece are equal to the number of divisions on stage micrometer. then after the calibration , multiply the eye piece divisions covered by cell or tissue being observed by the distance of each division say 2.5 micrometer... ( I am also not fully clear about it) u need to this all by your hands practice it. i don't like slide questions

Actually, your diagram should be proportionate to the specimen on the slide. Use the graticule to measure the length and then the width of the specimen under the microscope. And then using the same proportion you should draw your diagram. For e.g if the width of the specimen is .5mm and the length is one mm. Then your diagrams width should be 2.5 cm and length 10 cm. That is in the same proportion. So if you have a width of 5 cm, your length should be 20 cm and so on.
Hope this helped. :)

I think thickness refers to width and the length. You don't have a 3D microscope to measure the actual thickness. Since, its usually a transverse section, the length is actually the thickness of the tissue. So the thickness in the drawing should be relative to that in the microscope.

hmm....hamid there are sum questions in which they are asking to use high power and draw 4 or five cells......so any important tip for that

Not really. Just use the proportion rule in that too. Make sure your lines are clean and its not a sketch. The lines should be unbroken and drawn in one go. Do not shade. Since, its a high power drawing, you may see the nucleus or other organelles so just draw them. The best tip: Draw what you see.
